Stereo jack cable vs mono - what's the difference?

And as for whether it matters...

If you're hooking up a guitar to an amp, it doesn't matter at all. A stereo cable can be used.

However, if you're hooking up something that wants a stereo cable, you'll only get signal to one side if you use a mono cable.

A-ha... so which one is better, sound-wise, for guitar to effect or amp?

It makes no difference at all, unless some of the components in your signal chain are balanced.

Between the guitar and amp, you can use either, but there's no point using a TRS because the second + conductor just won't be used.

Most effects loops aren't balanced, so you wouldn't need a TRS cable there either.

The only place I've got TRS cables are in my recording rig running between my audio interface and my monitors, and that's only because both the audio interface and the monitors are balanced, so using the TRS actually cuts down on some noise.
