Completely possible, though tricker to line up than a press. And buying a decent drill that will make that hole for you comfortably would be just as much as a used drill press anyways.

Make sure you get one with a 1/2" chuck, the 3/8" wont cut it for big stuff like 3PDT's. I have and recomend the following:

A press will be nicer, but a hand drill will be more versatile for other projects, and easier to store and take care of. The hard part isn't really drilling, its making your marks accuratly. Many enclosures have rounded edges and that makes it tough to get exact measurements if you want two holes to be even. Having a press wont make them any straighter. If you use a center punch right, you wont drift with a hand drill. Get an automatic center punch like this:

Another nice thing about the DeWalt I linked is that it is two speed - for drilling you typically want slower with steady easy presure to avoid splashback that you will need to debur. If you do have that happen, its nice to have the faster setting so you can plug in a sanding bit and file that crap right off.
Also it has two batteries and they are NiCad, not Lithium, which I prefer. Whaaaaaaat you say? Yes I prefer NiCad because they don't slowly die even when not in use, like a Lithium would.
Also a hand drill is portable so you can bring it with you to your gigs and install that new set of sperzels on your guitar and make a pedal before the show that you forgot to bring. Think about that eh?
